Every Romulus shrine is marked on the map with a Wolf Icon. This shrine can be found in the Antico Ditrict in front of “Piramide Cestia” (The landmark). It is an easy shrine and can be done in less than 8 minutes. This trophy will pop once you loot the Scroll of Romulus and exit through the door.

While Da Vinci Disappearance takes place between Sequence 8 and 9, there isn't really the opportunity to play it then. However, from a story perspective, it actually makes sense to play it after the main story because they are 'recovered memories

The Florentine Noble attire was worn by Ezio Auditore in Florence during the beginning of Assassin's Creed II, before acquiring his father 's Assassin's Robes. In Assassin's Creed: Brotherhood, it is worn during the first repressed Cristina memory. The outfit is made accessible before the Apple of Eden memories when leaving the Animus.
Also @AngryFingers the reason it wasn't in Assassin's Creed 2, was because they were "repressed" memories and Desmond wasn't synch'd enough with Ezio to unlock those repressed memories. Or at least that's how I understood it.
The Da Vinci Disappearance (Brotherhood) is quite good; it makes the segue into Revelations smoother. If you liked the Desmond Files in Revelations, get the Lost Archive. The DLC for ACII is alright; its story is interesting and is referenced in Brotherhood, but the gameplay is more of the same, not too innovative.
